摘要
编码牛免疫缺陷病毒(BIV)穿膜蛋白的一个400bp的基因片断被克隆到pATH表达载体上,此重组的融合蛋白TrpE-Env8在大肠杆菌中得到高效表达,而且包含有与BIV抗体特异性反应的抗原决定簇.我们用此融合蛋白免疫balb/c,小鼠得到与BIV穿膜蛋白特异性反应的杂交瘤.此单克隆抗体在蛋白印迹法(westernblotting)分析中与重组的TrPE-Env8有特异性反应,用免疫酶染色法可检测BIV在体外的复制.
A 400bp gene segment encoding a partial peptide of Bovine Immunodeficiency Virus (BIV)transmembrane protein was cloned into pATH expression vector. The recombinant fusion protein trpE-Env8 expressed at a high level in E. colt and containing a major immunoreactive epitope. The fusion protein was used to immunize balb/c mice to generate hybridoma specifically against BIV transmembrane protein. The Mab was specific for the TrpE-Env8 protein in western blotting assay and detected specifically the BIV replication in vitro by immunoperoxidase assay.
