摘要
用体内自发突变,卡那霉素抗性筛选和体外限制酶切富集方法消除载体pNG35中新霉素磷酸转移酶基因内的限制酶位点HindⅢ,除去HindⅢ位点后的载体PNG35△H赋予宿主细胞的卡那霉素抗性水平远远高于原载体pNG35.
Author describes the method and result of removing Hind Ⅲ site in the neomycin phosphotransferase (NPT--I) gene through spontaneous mutation in vivo, kanamycin --selection and restriction enzyme digestion--enriching mutant in vitro. A plasmid pNG35△H without Hind Ⅲ site in NPT--I gene was obtained. This plasmid confers host cells much higher kanamycin--resistance level than the plasmid pNG35.
