流式细胞仪检测人脾LAK细胞

摘要 目的:探求LAK细胞对白血病疗效的监测手段。方法:用流式细胞仪测定5例成人脾淋巴因子激活的杀伤细胞(LAK细胞)和21例正常人外周血淋巴细胞的DNA指数(DI)及增殖周期。结果:人脾LAK细胞的增殖活力明显高于未激活的脾淋巴细胞和外周血淋巴细胞。培养(1~14d)期间人脾LAK细胞的增殖活力高,并且稳定。在人脾LAK细胞中,未找出非整倍体细胞,DI指数在正常范围。结论:流式细胞仪可用于判断人脾LAK细胞对白血病的疗效。 This research was aimed to find out a measure for observing the effect of lymphokine-activated killer (LAK ) cells on leukemia. METHODS: The DNA index (DI) and preliferative cycle of splenic LAK cells from 5 adults and peripheral lymphocytes from 21 normal subjects were estimated with flow cytometry (FCM ). RESULTS: The proliferative activity of splenic LAK cells was significantly higher than that of splenic cells and peripheral lymphocytes. It was highly active and stable during culture for a period of 1-14 d. Non-ploid cells were not found in the splenic LAK cells and the DI remained in normal range. CONCLUSIONS:FCM may be used to monitor the effect of allogenic LAK cells on leukemia.
