摘要
目的观察磷脂酰肌醇3位羟基激酶(PI-3K)的特异性阻断剂LY294002对阿魏酸钠抗β淀粉样蛋白(25-35)(Aβ25-35)诱导培养的皮层神经元凋亡作用的影响。探讨磷脂酰肌醇3位羟基激酶/Akt(PI-3K/Akt)信号转导通路是否参与了阿魏酸钠的抗凋亡作用。方法以Aβ25-35诱导大鼠皮层神经元凋亡模型,用倒置显微镜观察细胞形态变化,用Hoechst33258核染色分析神经元凋亡,并计算神经细胞的凋亡率。结果阿魏酸钠对Aβ25-35诱导的皮层神经元凋亡有保护作用,使神经细胞凋亡率下降。LY294002可以抑制阿魏酸钠的抗凋亡作用。结论在Aβ25-35诱导大鼠皮层神经元凋亡模型中,PI-3K/Akt信号转导通路参与了阿魏酸钠的抗凋亡作用。
Objective To observe the effects of a special inhibitor of phosphatidylinositol 3 - kinase ( PI - 3K) - LY294002 on sodium ferulate in the protection of cortical neurons against β- amyloid peptide (25 - 35 ) - induced apoptosis in vitro, To explore whether phosphatidylinositol 3 - kinase/Akt signal pathway is involved in anti - apoptosis action produced by sodium ferulate and its mechanism. Methods Apoptosis was induced by β - amyloid peptide (25 - 35) in cultured cortical neurons. Morphology of neurons was observed by microcopy and Hoeehst 33258 nucleus staining The percent of apoptosis was calculated. Results Sodium ferulate protected cultured cortical neurons from apoptosis induced by β - amyloid peptide (25 - 35). The neuronal apoptosis was reduced. LY294002 partly inhibited sodium ferulate protection. Conclusions Phosphatidylinositol 3 - kinase/AKT signal pathway is involved in anti - apoptosis action produced by sodium ferulate in β - amyloid peptide (25 - 35 ) - induced cortical neurons apoptosis.
