摘要
目的探讨高三酰甘油(TG)血症与肝脂酶(HL)启动子710T/C基因多态性的关系。方法运用聚合酶链反应-限制性内切酶片段长度多态性(PCR-RFLP)技术对112名正常TG者和103例高TG者的HL启动子710T/C基因多态性进行研究。结果HL启动子710C等位基因频率在高TG血症组(42.86%)与正常TG组(49.51%)中差异无显著性(P>0.05)。高TG组各基因型的各项血脂指标与正常TG组相同基因型的血脂指标比较,除低密度脂蛋白胆固醇(LDL-C)外差异均有显著性(P<0.05)。高TG女性组TT型、TC型的各项血脂指标与正常TG组相同基因型的血脂指标比较,差异均有显著性(P<0.05);在CC型中除总胆固醇(TC)、高密度脂蛋白胆固醇(HDL-C)外,其他血脂指标也与正常TG组相同基因型的血脂指标有差异(P<0.05)。在正常TG男性组中可见CC型的HDL-C浓度有增高趋势且明显高于TT型(P<0.05);在高TG男性组中,CC型的LDL-C浓度明显高于TT型和TC型,载脂蛋白A1(ApoA1)则明显降低(P<0.05)。未发现C等位基因是高TG发生的独立危险因素。结论肝脂酶启动子710T/C多态性与高TG血症的发生相关,并可影响血浆脂类的代谢,但不是一独立的危险因素。
Objective To investigate the relationship between hypertriglyceridemia and hepatic lipase promoter 710T/C polymorphism. Methods The hepatic lipase promoter 710T/C polymorphism of 103 hypertriglyceridemics and 112 healthy subjects were analyzed by polymerase chain reaction-restriction fragment length polymorphism. Results The frequencies of hepatic lipase promoter 710C were 42.86% and 49.51% in hypertriglyceridemics and in healthy subjects respectively. There was no significant difference ( P 〉 0.05 ). The hypertriglyceridemia various genotypes each item of lipid levels and the same gender healthy subjects had significant difference besides low-density lipoprotein cholesterol ( LDL-C ) ( P 〈 0.05 ). The hypertriglyceridemia female group TT, total cholesterol ( TC ) each item of lipid levels and the healthy subjects of same genotype lipid levels had significant difference besides LDL-C ( P 〈 0.05 ). CC genotype except TC and high-density lipoprotein cholesterol (HDL-C), other lipid levels also had the difference with healthy subjects same genotype lipid levels (P 〈0.05 ). The CC genotype HDL-C in healthy male subjects group had the tendency to increase and was higher than TT (P 〈0.05 ). In hypertriglyceridemia male group, the CC genotype LDL-C was obviously higher than TT and TC, apolipopretein A1 ( ApoA1 ) was obviously reduced ( P 〈 0.05 ). We have not discovered that the C allele was the independent risk factor of hyper triglyceridemia. Conclusions There was association between hepatic lipase promoter 710T/C polymorphism and hypertriglyceridemics in lipoprotein metabolism. But the C allele may not be a risk factor predictor.
