摘要
利用IPTG诱导表达含有猪圆环病毒2型(PCV2)Cap蛋白羧基端421-37nt的重组大肠杆菌pGEX-PCV421-37,经聚丙烯酰胺凝胶电泳(SDS-PAGE)粗分离,免疫BALB/c小鼠,利用PCV2感染PK15细胞筛选PCV2特异性单克隆抗体。共获得5株,腹水的IFA效价范围为1∶1600～1∶8000;除单抗26H5-A3为IgG1亚型外,32A8-G5、33G10-F4、12B5-G3和14E12-A6全部为IgM型;全部轻链都属κ型。
The recombinant bacteria pGEX-PCV421-37 which had the protein of Cap with 421-37nt in carboxyl terminus of PCV2 was expressed by using IPTG. Then SDS-PAGE electrophoresis was made to recover the protein by cutting gel method and the protein was immune to BALB/c mouse for monoclonal antibody production. We identified the five mAbs by IFA with PCV2 infected PK-15.The IFA titers of these mAbs were 1：1600-1：8000,in these mAbs 26H5-A3 was IgG1 and 32A8-G5, 33G10-F4、12B5-G3 and 14E12-A6 were IgM, all the light chain of five mAbs were κ.
