2-硝基咪唑-卟啉-二乙撑三氨四乙酸对小鼠Lewis肺癌的放射增敏效应 被引量:5

摘要 目的采用动物肿瘤模型探讨2硝基咪唑卟啉二乙撑三氨四乙酸(MISO卟啉DTTA)的放射增敏效应。方法将移植有Lewis肺癌的C57BL/6小鼠随机分为7个组:空白对照组、乏氧对照组(阻断小鼠荷瘤肢体血流10min)、单纯用药组(分为8.58、17.15mg/kg体重2个亚组腹腔内单次注射给药)、单纯放射组(分为7.5、15.0、25.0Gy3个亚组单次照射)、乏氧放射组(乏氧条件下分为7.5、15.0、25.0Gy3个亚组单次照射)、放射+药物组(分6个亚组)、乏氧放射+药物组(分6个亚组)。每组8只小鼠。采用肿瘤生长延缓方法进行放射增敏实验。结果当肿瘤体积达到实验开始时体积4倍时,分别测得8.58、17.15mg/kg体重的MISO卟啉DTTA在有氧状态下的放射增敏比平均为1.62和1.89,在乏氧状态下的平均为1.57和1.77。空白对照组、乏氧对照组、单纯用药1组和单纯用药2组的平均存活时间分别为(31.8+4.6)、(29.1+4.3)、(29.5+3.8)、(33.4+5.1)d(F=1.63,P=0.205)。结论8.58、17.15mg/kg体重的MISO卟啉DTTA对Lewis肺癌具有放射增敏效应,且对小鼠无毒性。 Objective To investigate the radiation-sensitizing effect of MISO-porphyrin-DTTA on animal model. Methods C57BL/6 mice transplanted with Lewis lung cancer were divided into 7 groups randomly: control; hypoxia alone; MISO-porphyrin-DTTA alone; radiation alone; radiation + MISO-porphyrin-DTTA; hypoxic radiation; and hypoxic radiation + MISO-porphyrin-DTTA. For hypoxia alone, the blood flow in the tumor beating leg was interrupted for 10 min. MISO-porphyrin-DTTA alone group was further divided into 2 sub-groups according to the dose of MISO-porphyrin-DTTA with 8.58 and 17.15 mg/kg, both in single intraperitoneal injection. Radiation alone group was divided into 3 sub-groups according to different radiation single dose (7.5, 15.0, 25.0 Gy). Radiation + MISO-porphyrin-DTTA group was divided into 6 sub-groups. Hypoxic radiation + MISO-porphyrin-DTTA group was divided into 6 sub-groups,each group including 8 mice. Method of tumor growth delay was used in the experiment. Results Mean sensitizing enhancer ratio on oxygen was 1.62 and 1.89, with 1.57 and 1.77 on hypoxia, according to 8.58 or 17.15 mg/kg MISO-porphyrin-DTTA when the tumor volume was 4 times of its initial volume. Conclusion MISO-porphyrin-DTTA on dose of 8.58 or 17.15 mg/kg do have radiation-sensitizing effect on Lewis lung cancer and are avirulent to C57BL/6 mice.
