摘要
目的评价椒蒿有效成分对心肌细胞的保护作用。方法采用SD大鼠乳鼠心肌细胞进行原代心肌细胞培养,然后行药物对心肌细胞最大无毒剂量的选择试验、药物在细胞外对CVB3 的直接灭活试验、不同药物浓度和不同作用时间对心肌细胞保护作用的试验。结果药物在1:8- 1:256稀释范围内对心肌细胞的生长无明显影响,<1:8时则出现药物毒性反应;药物与病毒作用后, 病毒滴度与对照组比较差异无统计学意义(P>0.05);药物在1:10-1:80的稀释范围内随着稀释度增加,其对心肌细胞的保护率增加至最高。随后稀释至1:160-1:320时则逐渐下降;药物作用于心肌细胞48 h后可明显降低CVB3在心肌细胞上的TCID_(50)。结论椒蒿可能是一种应用于临床治疗病毒性心肌炎急性期的较为理想药物。
Objective To evaluate the protective effects of JiaoHao extract (JHE) on myoeardium cells of rat with viral myocardifis(VMC). Methods VMC model in SD rats were infected by CVB3. VMC of SD rats were cultured in vitro and added JHE by different concentration and different times into the medium. Counting the number of VMC and recording the growth curve of cell in the JHE liquids. Cardiac cell proliferation was assessed by colorimetric assay. The largest non-toxic dosage of drug test was selected and the direct killing activity test was measured on CVB3 in vitrro, and protecting rate was calculated, too. Results From 1 : 8 to 1 : 256 dilute degrees in the range, the growth of myocardium cell was no change significantly. There was no difference between CVB3 group and CVB3 + drug group in the level of LgTCID50. The protecting rate was 56.9%, 68.8%, 75%, 95%, 44%, 23% respectively in the range of 1:10, 1:20, 1:40, 1:80, 1:160, 1:320 dilute degrees. Compared with CVB3 group,from 48 hours after being treated with JHE, the level of LgTCID50 in drug group was significantly decreased. Canclusion Our study indicates that JHE may be an efficient drugs for treating in acute phase of viral myocarditis.
