高效液相色谱法鉴定葡萄饮品质量及OPC含量测定

摘要 采用葡萄饮品试样的色谱图中出现的特征色谱峰与标准样品或参比样中出现的特征色谱峰相比较的方法实现对试样质量的鉴定。测定试样中OPC含量时,用C18(ODS)色谱柱(250 mm×4.6 mm,5μm)进行反相色谱分离。用体积比为13比87比0.05的乙腈+水十三氟乙酸的混合物作流动相,在定性及定量分析中采用紫外检测器,对方法的精密度与回收率作了试验,测定结果的RSD值在1.3%~1.7%,回收率在91%~116%之间,同时用传统的比色法对4种试样中的OPC含量作了比对分析,结果表明HPLC法的结果可信度更高。 Qualification of grape juice drinks was performed by comparing the characteristic chromatographic peaks in the chromatogram of the sample with those of the standard or reference sample. Based on the presence or not of those specific chromatographic peaks, judgement of the quality of sample was drawn. In the HPLC determination of contents of OPC, reverse-phase separation was carried out on a C18 (ODS) chromatographic column (250 mm×4. 6 mm, 5 μm). A mixture containing methyl cyanide, water and trifluoroacetic acid in the proportion of 13 to 87 to 0. 05 by volume was used as the mobile phase. UV-detector was used in both qualitative and quantitative analysis. Precision and recovery of this method were tested giving results of RSD's in the range of 1.3%-1.7% and of recoveries in the range of 91%- 116%. Validity of the proposed method was checked by a comparative analysis with the conventional colorimetric method, giving results of higher reliability.
