摘要
小牛软骨经盐酸胍抽提、丙酮分级沉淀及膜超滤等步骤获得了软骨抗肿瘤制剂(Cartitage Antiumor Preparation,CATP)。利用^3H-TdR参入,测定肿瘤干细胞及细胞死亡率等方法研究了CATP对肿瘤细胞及血管内皮细胞的换制效应,并对小鼠移植性肿瘤进行了抗肿瘤作用的研究。结果表明:(1)合适浓度的CATP可显著地抑制肿瘤细胞和血管内皮细胞的生长,而不抑制正常细胞;
Cartilage antitumor preparation (CATP) was isolated from a guanidine hydrochloride extract of bovine cartilage by acetone fractioned precipitation and ultrafiltration. By the methods of 3H-TdR incorporation, tumor stem cell assay and the determination of mortality of cell, the effect of CATP on the tumor cell lines and bovine artery endothelial cells were investigated and the antitumor effect of CATP was observed by the experiment of transplanation of tumor into mice. The results were as follows; (1) CATP had the effects of inhibiting angiogenesis and tumor cells; (2) CATP markedly inhibited both the growth of sc solid tumor of sarcoma 180 and that of Lewis lung carcinoma in the mice, inhibition rate was 50%-60% ; (3) Thymus weight of treated group obviously increased in comparison with control group, indicating that CATP had immunoregulatory effect. The experimental results furnished evidence in order to apply to clinic.
