摘要
目的:从细胞凋亡角度探讨当归多糖对豚鼠耳部银屑病样模型的作用。方法:50 g/L普萘洛尔乳剂外涂豚鼠耳背皮肤,制备银屑病样模型,然后随机分成模型对照组(Ⅰ)、低剂量组(Ⅱ)、中剂量组(Ⅲ)、高剂量组(Ⅳ),依次给予生理盐水、100,200,400 mg/kg当归多糖腹腔注射。应用末端标记技术(TUNEL)分别检测各组皮损治疗前后表皮角质形成细胞凋亡情况。结果:造模后豚鼠耳部上皮角质形成细胞凋亡指数与涂药前相比明显增加(P<0.05),Ⅱ、Ⅲ、Ⅳ组治疗后皮损凋亡指数显著高于治疗前(PⅡ<0.05,PⅢ<0.05,PⅣ<0.01)。结论:当归多糖对普萘洛尔诱发的豚鼠耳背银屑病样模型有治疗作用,其作用机理可能与其促进角质形成细胞的凋亡有关。
Objective: To investigate the influence of angelica polysaccharide on apoptosis in psoriasis- like lesions of guinea pigs. Methods: Propranolol emulsion (50 g/L) was applied on ears' back of Guinea pigs to produce the psoriasis-like models, then the forty guinea pigs randomly divided into four groups., model control group( Ⅰ ), low-dose group( Ⅱ ), mid-dose group( Ⅲ )and high-dose group(Ⅳ). Normal saline and 100, 200 and 400 mg/kg angelica polysaccharide were injected intra-abdominally respectively. And in each group the apoptosis of keratinocytes in their skin lesions before and after treatment was detected by terminal deoxynucleotidyl transferase-mediated dUTP nick end-labeling(TUNEL) technique. Results: There was a large number of apoptosis keratinocytes in psoriasis-like lesions of guinea pigs' ears. Compared with that of normal guinea pigs, the apoptosis index was increased(P〈0.05). Expression of apoptotic cells in psoriasis-like lesions after injecting angelica polysaccharide in group Ⅱ , Ⅲ and Ⅳ was significantly higher than that before treatments(PⅡ〈0.05 and PⅢ〈 0.05, PⅣ〈 0.01 ) respectively. Conclusion. Angelica polysaccharide might have a therapeutic effect on the psoriasis-like lesions in the epidermis of guinea pigs' ears induced by propranolol ointment, and the mechanism may be related to the increase in the frequency of apoptotic keratinocytes in lesions.
