摘要 目的:探讨原代培养人胎膜细胞中白细胞介素(IL-18)的表达及肿瘤坏死因子-α(TNF-α)对体外培养的胎膜细胞IL-18表达的调控及其在胎膜早破发生中的作用。方法:取16例剖宫产产妇胎膜,将胰蛋白酶消化而得到的胎膜细胞进行原代培养。采用ELISA法对TNF-α(20μg/L)刺激前后细胞培养液中IL-18的含量进行检测。结果:无TNF-α刺激的胎膜细胞IL-18表达为(235.74±29.59)μg/L,经TNF-α刺激后表达为(392.46±89.82)μg/L,表达量明显增高,差异有显著性(P<0.05)。结论:TNF-α刺激胎膜组织生成IL-18可能与胎膜早破密切相关,在胎膜早破的发生中可能起一定作用。 Objective: To detect the content of IL-18 in premature of membranes and to explore its relations with premature rupture of membranes (PROM). Methods: Fetal membranes (n=16), collected at the time of elective repeat cesarean section prior to labor from normal term gestation without complications of pregnancy, were placed in an organ explant system with recombinant tumor necrosis factor-alpha (TNF-α, 20 μg/L)for 24 hours. Tissue after stimulation was subjected to ELISA to study the expression of IL-18 in tissue culture supernatants. Results were analyzed using the t-test for paired samples. Results: ELISA showed induction of IL-18in human fetal membranes after TNF-a stimulation. IL-18 expression was constitutive in both TNF stimulated and control membranes. IL-18 was significantly higher in TNF-α stimulation E(235. 74 ± 29.59)μg/L vs (392.46±89.82)μg/L,P〈0. 05]. Conclusion: TNF-α may play an important role in premature rupture of membranes.
