Expression of Angiopoietin-2 and Its Clinical Significance in Colorectal Carcinoma

摘要 目的:研究血管生成素-2(Ang-2)蛋白在人大肠癌组织中的表达及其与大肠癌临床病理特征的关系。方法:应用免疫组化法检测40例大肠癌及其癌旁正常组织中Ang-2蛋白表达水平。结果:大肠癌组织中Ang-2蛋白表达阳性率77.5%(31/40),明显高于癌旁正常组织40%(16/40)(P<0.01);Ang-2表达水平与患者性别、年龄及癌组织的部位、大小、分化程度、淋巴结转移无关(P>0.05);隆起型大肠癌Ang-2蛋白表达水平高于溃疡型大肠癌(P<0.01);肠壁浸润越深,Ang-2蛋白表达水平越高(P<0.01);有血道转移的比无血道转移的Ang-2表达水平高(P<0.05);Dukes分期D期Ang-2蛋白表达水平明显高于A期(P<0.01)。结论:Ang-2蛋白表达水平与大肠癌组织大体形态、浸润深度、血道转移及Dukes分期有关,提示Ang-2蛋白可能促进了大肠癌的生长及转移。 Objective: To study the expression of Angiopoietin-2(Ang-2) protein in colorectal carcinoma and evaluate the relationship between Ang-2 protein level and clinical parameters. Methods: Expression of Ang-2 was examined in 40 colorectal carcinoma and adjacent normal tissues by immunohistochemical technique. Results: The positive rate of Ang-2 protein was significantly higher in colorectal carcinoma than in normal tissues (77.5% vs 40%, P〈0.01). Neither patients' gender nor age correlated with the expression level of Ang-2(both P〉0.05). No correlations were found between the expression level of Ang-2 and the site, size or histological differentiation of colorectal carcinoma(P〉0.05). The expression level of Ang-2 in colorectal carcinoma with lymph node metastasis was not different from that without lymph node metastasis(P〉0.05). The positive rate of Ang-2 protein was higher in projecting carcinoma of colorectum than in ulcerative carcinoma of colorectum(P〈0.01), was lower in colorectal carcinoma of Dukes'A stage than that of Dukes'D stage(P〈0.01), and was higher in colorectal carcinoma with metastasis from blood vessels than that without metastasis from blood vessels(P〈0. 05). The deeper extent of infiltrating, the higher, the Ang-2 positive rate was (P〈0.01). Conclusion: The expression level of Ang-2 protein might be related to the shape of colorectal carcinoma, invasion ability, blood vessel metastasis and Dukes' staging in colorectal carcinoma. It is suggested that high expression of Ang-2 protein may play an important role in the progression and metastasis of colorectal carcinoma.
