摘要
采用病变抑制法和^(125)I放射免疫法在3种细胞培养系统上测定了天花粉蛋白对7种病毒的抑制作用。结果表明,天花粉蛋白浓度在10μg/ml以下时,对细胞无明显的毒性作用,在0.001~10μg/ml时对乙型脑炎、柯萨奇B 2、麻疹、腺病毒3型、单纯疱疹病毒Ⅰ型、水泡性口炎病毒及乙型肝炎病毒等均有明显的抑制作用,并且这种抑制作用随天花粉蛋白的浓度增大而提高。浓度在0.01μ/ml时可以抑制由100TCID50病毒引起细胞病变的50%。本文提示,天花粉可能是继干扰素之后另一种具有广谱抗病毒作用的蛋白。
A study on the antiviral activitis of a purified protein ( TKP ) isolated from root of Trichosanthes kirilwii was carried out three cell culture systems using CPE inhibition method and 125I radioimmunoassay. The results obtained indicated that TKP at concentrations less than 10μg/ml did not appear to be toxic to three types of cell cultures. A significant concentration dependent inhibition of CPE caused by the viruses of measles, Japen-ese B encephalitis, coxackie B2, herpes simplex type 1, as well as adeno virus 3 and vesicular stomatitis virus in WISH or Vero cell cultures was observed by simultaneously adding I KP in concentrations of 0.001-10 μg/ml with virus challenge. As an example, TKP in concentration of 0.01 μg/ml may efficiently inhibit 50% of CPE caused by 100 TCID50 of Cox-B2.TKP was also shown to inhibit HBsAg secretion by 2. 2.15 cells which were generated by transfection with HBV DNA.These results suggest that FKP is a potential wide-spectum antiviral protein.
