摘要
烟草花叶病毒(TMV)外壳蛋白的N-端乙酰化是在翻译开始后立即进行的。本文以TMV外壳蛋白N-端二肽Ser—Tyr为底物、醋酸盐为转乙酰基供体的反应体系,证明烟草乙酰基转移酶能将Ser—Tyr转化成N—Ac—Ser—Tyr。烟草乙酰基转移酶的最适pH为5.0,需要Mg~^(++)及—SH激活。推论烟草乙酰基转移酶可能与TMV外壳蛋白N-端的乙酰化有关。
In 1958, Narita firstly found that the N-terminal of the coat protein of tobacco mosaic virus was blocked by an acetyl group. Now a lot of N-acyl -proteins has been reported. Filner ( 1974 ) reported that the process of the N-acylation of the coat protein of TMV was completed at the early stage of translation. But about the N-acyltransferase little was known. This paper describes the N-terminal dipeptide Ser-Tyr of the coat protein of TMV could be acylated by the N-acyltraasferase in the presence of the acetyl donor 3H-acetate, The N-Ac-Ser-Tyr and Ser-Tyr was separated with silica gel thin layer chromatography,developed with Pauly's reagent and scanned at 445 nm. The optimal pH of the acyltransferase is 5.0 and Mg++ and -SH are essential for the enzyme activity. It was deduced that acyltransferase from tobacco leaves may be responsible for the N-terminal acylation of the coat protein of TMV at the early stage of translation.
