摘要
根据基因组序列分析,对预测为鸭乙型肝炎病毒(DHBV)Pre-S的两段多肽(P37-49,P63-79)进行了合成.两段多肽分别与DHBs/Pre-S兔免疫血清做放射免疫沉淀反应,结果P37-49可部分被沉淀.用蛋白印迹-免疫酶染色法,以P37-49交联牛血清白蛋白免疫家兔获得的抗体,可与纯化的DHBs/Pre-S+S中分子量为35kD的多肽(DHBV Pre-S)起反应。证明这一肽段具有DHBVPre—S的一个抗原决定簇.
Based on the scqueices of cloned duck hepatitis B virus ( DHBV ) genome, two predicted possible determinants of DHBV Pre-S antigen were synthesized ( P37-49, P63-79 ) . These peptides when reacted with rabbit anti-DHBs/Pre-S by radioimmunoprecipitation showed partial precipitation with P37-49. P37-49 was cross-linked to bovine serum albumin for the preparation of antiserum. By Western blotting this antiserum reacted with a 35 kD peptide of purified DHBs/Pre S+S,which is recognized as containing the Pre-S of DHBV. Therefore, peptide P37-49 is one of the determinants of DHBV Pre-S antigen.
