摘要
目的检测促凋亡蛋白BAD在人乳腺导管增生及乳腺癌组织中表达,探讨其在乳腺癌癌变过程中的作用及意义。方法采用免疫组织化学S-P法,检测131例乳腺导管增生及乳腺癌组织中促凋亡蛋白BAD的表达。结果BAD蛋白在正常乳腺组织中的阳性率为33.3%(3/9)。从普通导管增生、轻中度不典型导管增生、重度不典型增生及导管原位癌到浸润性导管癌组织,BAD蛋白表达阳性率呈递增趋势,在乳腺普通导管增生和重度不典型增生及原位癌组间阳性表达率差异均具有统计学意义(P<0.05)。结论促凋亡蛋白BAD蛋白表达在乳腺癌癌变过程中呈递增趋势;BAD蛋白表达水平与组织学分级有关,组织学分级越低,其蛋白表达阳性率越高。
Objective To determine the expression of Bcl-xl/Bcl-2-associated death (BAD) protein in human mammary intraductal hyperplasia and breast carcinomas to explore the action and significance of BAD protein in their progression. Methods BAD expressions in 131 cases of mammary intraduetal hyperplasia and breast cancer were determined by S-P immunohistochemistry assay. Results The positive rate of BAD protein in normal mammary tissue was 33.3% (3/9), while that of BAD presented gradual asce.nding trend in the following order: normal mammary duet hyperplasia, mild and moderate atypical intraductal hyperplasia, severe atypical hyperplasia, intraductal in situ neoplasma and invasive intraductal carcinoma There were significant differences in positive expression of BAD among intraductal hyperplasia, severe atypical hyperplasia and in situ carcinoma (P = 0. 022). Conclusions BAD protein expresses in a gradual ascending trend in the development of breast cancer, the level of which could be related to histological grade, i.e. the low the histological grade, the high the positive rates of the BAD protein expression.
