大黄素脂质体对Hep-2细胞毒的检测 被引量:4

摘要 采用逆相蒸发法制备了大黄素脂质体,测定了脂质体的包封率和粒径.通过苔盼蓝拒染法检测脂质体对Hep2细胞的剂量和时间效应,初步研究了脂质体对Hep-2细胞的毒性,并根据加入脂质体后Hep-2细胞一周内的生长情况制作了生长曲线.结果测得由该法制备的大黄素脂质体平均包封率高达90.6%,透射电镜下观察到脂质体呈椭圆囊状结构,粒径分印均匀,其平均粒径分布在0.02~0.1μm之间,为单室脂质体.细胞毒实验表明大黄素脂质体对Hep-2细胞存在明最的细胞毒作用,且具有剂草和时间效应,随着脂质体中大黄素浓度从0.48mg·L^-1增加到14.48mg·L^-1,死细胞百分率由23.15%增至94.5%,并测得脂质体对Hep-2细胞48h的半数抑制浓度IC50。为4.43mg·L^-1,浓度为7.24mg·L^-1的脂质休勺Hep-2细胞作用12h后,死细胞百分率增加到95%以上,加脂质体的Hep-2细胞生长曲线与没有加脂质体的正常Hep-2细胞生长曲线比较,加了脂质体的Hep-2细胞的生长曲线呈明显下降趋势,说明大黄素脂质体对Hep-2细胞的生长存在抑制作用. Emodin liposome was prepared by the reverse evaporating method. The entrapment efficiency and particle size of the emodin liposome were determined. Through the examination of dose and timeeffect by trypan blue method,the toxicity of emdoin liposome in Hep 2 ceils was studied,and growth curve of liposome treated Hep 2 ceils was drawn. The results show that the emodin liposome prepared by the reverse evaporating method has a high entrapment efficiency of 90.6 %, and its particle size is 0.02-0.1 μm. The results of cytotoxieity study indicate that the liposome exhibits evident toxicity in Hep-2 ceils and has dose-and time-effect,as the concentration of emodin liposome raised from 0.48 mg·L^-1 to 14.48 mg·L^-1 , the ratio of dead cells increased from 23. 15% to 94.5% ,and 48 h IC50 of the liposome is 4.43 mg·L^-1 ,after 12 h liposome with a concentration of 7.24 mg·L^-1 was acted on Hep-2 cells,the ratio of dead cells increased to over 95%. Comparing the growth curve of liposome-treated ceils with normal Hep-2 ceils, the growth curve of liposome treated cells declined evidently and this result indicates that emodin liposome exhibits control effect on Hep- 2 ceils growth.
