摘要
目的观察肝细胞生长因子(HGF)对大鼠减体积肝移植术后再生的影响。方法建立大鼠减体积(60%)肝移植模型,将大鼠随机分为生理盐水对照组(A组)和肝细胞生长因子治疗组(B组),每组动物60只。受体于术后皮下注射给药(HGF,100mg/kg)每天一次,直至处死。观察术后1、2、3、5、7天受体残肝重/减体积前全肝重,肝细胞有丝分裂指数(MI)、增殖细胞核抗原标记指数(PCNA LI)和溴脱氧核苷尿嘧啶的掺入指数(BrdU LI)以及术后血清丙氨酸氨基转移酶(ATL)、天冬氨酸转氨酶(AST)水平。结果HGF能显著提高受体肝细胞MI、PCNA LI、BrdU LI及残肝重/减体积前全肝重(P<0.05,P<0.01),两组血清丙氨酸氨基转移酶(ALT)、天冬氨酸转氨酶(AST)水平无显著差异(P>0.05)。结论肝细胞生长因子能显著促进大鼠减体积肝移植术后肝组织的再生,而对术后肝脏功能无明显损害作用。
Objective To study the effects of HGF on liver regeneration after reduced-size liver transplantation in rats. Method 120 rats were divided into two groups,control(group A) and HGF(group B). After reduced-size(60%) liver transplantation,the recipients of group B were administrated with HGF(100mg/kg/d) until the animals were killed at 24,48, 72,120 and 168 hour after surgery. Mitotic index( MI), Proliferating Cell Nuclear Antigen Labeling index( PCNA LI), Bromodeoxyuridine Labeling index(BrdU LI)of hepatocytes and ALT,AST of serum were measured. Results HGF can significantly augmented Mi,PCNA LI and BrdU LI of hepatocytes after the transplantation(P〈50.05, P〈0.01 ). No significant differences of serum ALT and AST levels were observed between the two groups(P〉0. 05). Conclusion HGF improved liver regeneration after reduced-size liver transplantation in rats and has no negative effects on liver function.
