摘要
背景:六味安消胶囊能明显加快胃排空,促进肠推进运动。目的:探讨六味安消胶囊及其组分对豚鼠结肠P物质(SP)和黏蛋白2(MUC2)表达的影响。方法:20只豚鼠随机分为4组:正常对照组、大黄组、诃子组和六味安消胶囊组,后3组分别予相应药物灌服30天。处死动物,观察结肠黑变情况;对盲肠和近段结肠切片行SP、MUC2免疫组化染色和图像分析;对肌间神经丛铺片行嗜银染色,计数神经元细胞数。结果:大黄组结肠有黑变,以盲肠和近段结肠为著。大黄组、诃子组和六味安消胶囊组近段结肠SP免疫反应阳性纤维较正常对照组显著增多(阳性面积比:6.2%±1.7%、5.3%±0.8%和5.4%±0.9%对2.1%±0.7%,P<0.01),免疫阳性增强(灰度:131.50±1.16、133.79±0.63和133.78±0.67对146.20±2.03,P<0.01),大黄组近段结肠SP免疫阳性又显著强于诃子组和六味安消胶囊组(P<0.05)。各组盲肠和近段结肠MUC2的表达无明显差异。大黄组肌间神经丛神经元细胞数显著低于其余各组(11.60±1.52对17.00±1.87、18.60±3.78和17.80±2.95,P<0.05)。结论:六味安消胶囊可能通过其主药大黄改变肠道内SP含量而发挥消积导滞作用。六味安消胶囊和大黄对结肠MUC2的表达无明显影响。
Background: Liuwei Anxiao Capsule can speed up gastric emptying and intestinal propulsion. Ahns: To study the effects of Liuwei Anxiao Capsule and its ingredients on the colonic expression of substance P (SP) and mucin 2 (MUC2) in guinea pigs. Methods: Twenty guinea pigs were randomly divided into 4 groups: normal control group, rhubarb group, fructus group, and Liuwei Anxiao Capsule group. The latter 3 groups received a daily intragastric administration of the respective drugs for 30 days. The animals were then killed to observe the nigrities of colonic mucosa. Immunohistochemistry and image analysis were used to investigate the distribution of SP and MUC2 in sections of cecum and proximal colon. Silver staining was performed on stretched preparation of myenteric plexus and positive nerve cells were counted. Results: Macroscopically, the mucosa of colon, in particular cecum and proximal colon was brown in color in rhubarb group. In rhubarb group, fructus group and Liuwei Anxiao Capsule group, the SP positive fibers of proximal colon were increased in number more than those in normal control group (positive area ratio: 6.2%±1.7%, 5.3%±0.8%, and 5.4%±0.9% vs. 2.1%±0.7%, P〈0.01). The degree of immunostaining was obviously more intense (gray scale: 131.50±1.16, 133.79±0.63, and 133.78±0.67 vs. 146.20±2.03, P〈0.01); in the rhubarb group the immunostaining degree of SP positive fibers of proximal colon was more intense than that in the fructus group and Liuwei Anxiao Capsule group (P〈0.05). There was no difference in the expression of MUC2 at cecum and proximal colonic mucosa between the groups. Silver stained nerve cells of myenteric plexus in the rhubarb group were decreased significantly than those in the normal control group, fructus group and Liuwei Anxiao Capsule group (11.60±1.52 vs. 17.00±1.87, 18.60±3.78, and 17.80±2.95, P〈0.05). Conclusions: Liuwei Anxiao Capsule may promote intestinal transit through its principal ingredient rhubarb, which increases the colonic concentration of SP. Liuwei Anxiao Capsule and rhubarb do not influence the colonic expression of MUC2.
