Establishment of miniature pig model of chronic ischemic heart disease

摘要 目的:通过改良手术方法建立稳定、安全的小型猪慢性缺血性心脏病模型,并建立活体判断模型建成与否的观察指标。方法:手术于2005-01/10在香港大学动物实验中心进行。取健康巴马小型猪17只,左侧开胸后应用改良法(两线平移牵引法)在小型猪左冠状动脉回旋支起始部放置Ameroid环,具体改良环节为:①用两条丝线牵住冠状动脉的左回旋支并将两线之间的左回旋支与Ameroid环的缺槽平行对齐,立即将两者同时做相反方向的运动,迅速将其套入Ameroid环内。②在给小型猪做经口行气管插管时,气管插管在过了喉头后应顺着这个弯曲向背侧方向进入气管。4周后进行活体冠状动脉造影。结果:17只猪中有16只猪术后存活8周以上,活体冠状动脉造影显示左冠状动脉回旋支均闭塞。结论:通过改良手术方法能建立起稳定、安全的小型猪慢性缺血性心脏病模型,用冠状造影在活体上可确定模型成功与否。 AIM: To explore a more safe and stable pig model of chronic ischemic heart disease by improving the operation method, and establish a living observation index to judge whether the model is established successfully. METHODS: The experiment was conducted in the animal experimental center of the University of Hong Kong between January and October 2005. Totally 17 healthy Bama miniature pigs were selected to perform the thoracotomy and an Ameroid constrictor was placed on the initial portion of left circumflex branch of coronary artery by the improved operation method (amphi-thread translation draught method): ①The left circumflex branch of coronary artery was drafted with two threads by translation to be parallel to the gap of the Ameroid constrictor, then the left circumflex branch was put into the constrictos with the quick negative direction movement. ②The tube was passed the throat and inserted into the trachea through the incurvation, when conducting the endotracheal intubation. Four weeks later, coronary arteriography was done on living body. RESULTS: Totally 16 pigs of the 17 ones survived more than 8 weeks. Complete occlusion of left circumflex coronary artery was observed by coronary arteriography. CONCLUSION: A more safe and stable pig model can be established by the improved operation method, and the coronary arteriography on living body can judge whether the model is established successfully.
