卵泡刺激素受体表达与卵巢肿瘤发病的关系 被引量:4

摘要 目的:探讨卵泡刺激素受体与上皮性卵巢肿瘤的关系。方法:应用半定量逆转录-聚合酶链式反应(RT-PCR),免疫印迹(Westernblot)检测10例对照组,30例良性组及32例恶性组的卵泡刺激素受体(FSHR)mRNA和蛋白的表达情况。结果:FSHRmRNA在对照组中均表达(10/10),良性组中表达率为96.7%(29/30),恶性组中表达率为59.4%(19/32);其蛋白在对照组中均表达(10/10),良性组中表达率为90%(27/30),恶性组中表达率为46.9%(15/32),恶性组中FSHRmRNA及蛋白表达率明显低于对照组和良性组(均P<0.01),而且FSHR的表达程度,从对照组到良性组再到恶性组呈下降趋势。结论:在卵巢癌组织中卵泡刺激素受体的表达率及表达程度显著降低,卵巢癌细胞的增殖并非完全依赖促性腺激素-卵泡刺激素。 Objective: To investigate the relationship of follicle-stimulating hormone receptor (FSHR) and epithelial ovarian cancer. Methods: Ten normal ovaries, thirty benign ovarian tumors, thirty-two epithelial ovarian carcinomas were examined for FSHR mRNA and protein expression by semi-quatitive reverse transcription polymerase reaction (RT-PCR) and Western blot. Results: Expression of FSHR mRNA was found in 100%(10/10) of normal ovaries, 97.6% (29/30) of benign ovarian tumors, 59.4 % (19/32) of epithelial ovarian carcinoma. On the other hand, the protein expression in three tissues was 100% (10/10), 90% (27/30) and 46.9% (15/32)respectively .The expression rate of FSHR mRNA and protein in ovarian carcinoma tissue was significantly lower than that in the normal ovarian tissue and benign ovarian tumors (P 〈 0.01). What is more,the expression degree of FSHR had a decrease tendency in order from malignant, normal ovarian tissue to the epithelial ovarian carcinoma. Conclusion: The expression rate and degree of FSHR in the cpithelial ovarian cancer tissue significantly decrease,the proliferation of ovarian cancer cells is not thoroughly dependent on gonadotropic hormone-follicle stimulating hormone.
