摘要
用日龄2天的SD大鼠大脑皮层进行分散神经细胞原代培养,通过氰化物阻断细胞呼吸链的方法,模拟了大脑皮层神经细胞的急性缺氧损害模型。研究了氟桂嗪对缺氧损害细胞的作用,观察到缺氧细胞恢复过程中3H─TdR和3H─UR的参入量显著高于对照组;氟桂嗪保护组细胞3H─TdR和3H一UR参入量与缺氧组相比显著降低;氟桂嗪也可使正常皮层细胞3H─TdR和3H─UR参入量减少。
Primary cultures of rat cerebral cortical neurons were prepared from cortices of 2 days Sprague──Dawley rats and made an attempt to imitate the hypoxic neurons injury for testing neuroprotective effect of flunarizine. Hypoxia was induced hy adding lmmol/L sodium cyanide to the incubation medium of the neurons. It was found that hypoxic group had significantly higher incorporation of 3H-TdR and 3H-UR than control group, while flunarizine can decrease the incorporation of 3H-TdR and 3H-UR. In addition, flunarizine can decrease the incorporation of 3H-TDR and 3H-UR in normal cultured neurons. The results showed that flunarizine can regulate the synthesis of DNA amd RNA
