摘要
目的探讨CD105在胃癌新生血管中的表达及其作为肿瘤新生血管标记物的临床价值。方法采用鼠抗人CD105单克隆抗体(M ab),应用免疫组织化学技术检测80例胃癌手术切除标本及80例对照组织的微血管密度(MVD),并以血管内皮生长因子(VEGF)的表达作为对照研究。结果MVD及VEGF在胃癌组织与对照组织间表达的差异有显著性(P<0.001,P<0.01);VEGF在胃癌组织中的表达与CD105的表达呈显著正相关(P<0.01);胃癌组织MVD和VEGF的表达均与胃癌的淋巴结转移、远处转移及Dukes分期有关(P<0.01)。结论CD105在胃癌组织新生血管有良好的表达,可以作为胃癌新生血管有价值的标记物。以CD105测得的MVD可以被认为是与肿瘤生长、转移及预后有关的一种重要指标。
Objective To explore expression and clinical significance of CD105 and the vascular endothelial growth factor(VEGF) as a marker of neoplasm-related angiogenesis in stomach cancer. Methods Immunohistochemical staining with anti-CD105 monoclonal antibody(MaD) and anti- VEGF MaD was used to determine CD105 expression and the microvessel density (MVD) in tissue specimens from 80 cases of stomach cancer and 80 normal control specimens. Results The MVD marked by anti-CD105 and the positivity of VEGF in stomach cancer tissues were significantly higher than those in control tissues (P 〈 0.001, P 〈0. 01 ). VEGF expression was positively correlated with CD105 expression (P 〈 0. 01 ). MVD expressing CD105 and the expression of VEGF were closely associated with the lymph node metastasis, distant metastasis and Dukes stages of stomach cancer( P 〈 0. 01 ). Conclusions CD105 is expressed highly in the vascular endothelia of microvessels of stomach cancer, and its expression can be used as a valuable marker of angiogenesis in stomach cancer. The MVD marked by anti-CD105 is associated with tumor growth, metastasis and prognosis.
