摘要
目的探讨体外诱导骨髓基质干细胞(MSCs)形成组织工程化软骨的条件。方法经体外原代及传代培养,获取骨髓基质干细胞,高密度传代培养第3代诱导MSCs分化为软骨样细胞,以纤维蛋白粘合剂(FS)为载体,两者混合培养形成组织工程化软骨植入物(细胞浓度为5×106/m l),观察细胞的形态及生长特点。结果骨髓基质干细胞具有明显的增殖能力,贴壁率高,能诱导分化为软骨样细胞,形成甲苯胺蓝异染的软骨基质。结论骨髓基质干细胞可诱导分化为软骨样细胞,并能形成组织工程化软骨组织。
Objective To investigate the culture conditions of bone marrow stromal cells (MSCs) to form the tissue engineering cartilage. Methods The MSCs were obtained by the method of primary and passage culture in vitro. Chondrocytes derived from MSCs were obtained with high initial cell density subculture. The cells were cultured and transferred to the 3rd generation, and mixed with fibrin sealing (FS,5 × 10^6cells/ml). The conformation and growing feature of the tissue engineering cartilage were observed. Results The cultivated MSCs had shown evident reproductive activity and high rate of adherence. The histological results showed that cartilage matrix was stained positively for toluidine blue. Conclusion Chondrocytes can be derived from MSCs and form into the tissue engireering cartilage with FS.
