摘要
以SDS正丁醇-正庚烷-水组成的微乳体系作为展开剂,通过聚酰胺薄层层析,探讨不同类型微乳液对葛根异黄酮分离检测效果的影响。结果表明,选择含水量为70%的微乳液作为展开剂,分离效果明显,一次检测出12个黄酮化合物;与以三氯甲烷-甲醇-水(7:2.5:0.25)为展开剂的硅胶薄层相比,微乳薄层色谱对葛根异黄酮的检测效果和灵敏度显著提高,为实验研究中常规检测提供了简便快捷的方法。
The study adopted polyamide thin layer chromatography with different sodium dodecyle suphate/n-C4H9OH/n-C7H16/H2O micro-emulsions as its mobile phase to isolate and detect isoflavonoids in Pueraria. The results indicated that polyamide thin layer chromatography with the micro-emulsion whose water content was 70% used as the mobile phase performed best in isoflavonoids isolation and detection of pueraria and it could detect twelve isoflavonoids one time ;compared with silica gel thin layer chromatography with chloroform-methanol-water (7 : 2.5 : 0.25) as its mobile,polyamide thin layer chromatography with microemulsion as its mobile phase presented a significantly increased sensitivity and thus it could be used as one convenient and fast way in the routine detection in experiment and research.
