摘要 构建抗白血病转移药物筛选模型。K562细胞转染GFP质粒后,经筛选,将稳定高表达GFP的K562细胞注射到鸡胚里,孵化后观察K562细胞在鸡胚中转移情况。在其脑部、胸骨及心脏可看到明显的荧光,最频繁发生的部位是脑部。成功构建含GFP的白血病鸡胚模型,并验证了虎眼万年青皂甙(OSW-1)和链激酶(SK)联合用药的抗转移效果。 An imageable fluorescent chick-embryo metastatic cancer model was established with the use of green fluorescent protein (GFP). K562 cells which were transfected with GFP-N2 and screened were injected in the chick embryo. GFP-K562 carcinoma metastases were visualized by fluorescence, and GFP-K562 was visualized in the brain and sternum of the developing chick embryo, with the most frequent site being the brain. The combination of streptokinase and OSW-1 has been evaluated in the GFP metastatic model.
