摘要
目的:观察大鼠脊髓损伤后应用盐酸川芎嗪注射液对诱导型一氧化氮合酶表达及细胞凋亡的影响。方法:实验于2005-07/2005-11在华中科技大学同济医学院附属协和医院骨科实验室完成。采用Allen’s法将72只成年SD大鼠造成脊髓中度损伤模型,随机分为两组:损伤组和川芎嗪组,分别给予生理盐水及川芎嗪治疗,损伤后不同时间点(8h,1d,3d,1周,2周,3周)处死大鼠,用苏木精-伊红染色观察损伤脊髓组织病理变化,用免疫组化染色检测诱导型一氧化氮合酶阳性细胞,原位末端标记法标记凋亡细胞。观察各组大鼠各时间点组织学检查、诱导型一氧化氮合酶表达阳性细胞率和凋亡指数。结果:72只大鼠全部进入结果分析。①川芎嗪组大鼠与损伤组比较,脊髓出血明显减少,可见点灶状出血、坏死,范围较局限,神经细胞肿胀较轻,组织水肿较轻,空泡变性较少。②川芎嗪组与损伤组均发现诱导型一氧化氮合酶表达阳性细胞,可见于神经元、神经胶质细胞、血管内皮细胞、室管膜细胞,均在1周时达高峰,在8h,1d,3d,1周,2周,3周的时间点间进行阳性细胞率比较,差异均有显著性意义(t=2.234~13.412,P<0.05)。③川芎嗪组与损伤组均发现凋亡细胞,1周达高峰,在8h,1d,3d,1周,2周,3周时间点间进行细胞凋亡指数比较,差异均有显著性意义(t=2.417~3.689,P<0.05)。结论:川芎嗪注射液能抑制脊髓损伤后诱导型一氧化氮合酶表达及神经细胞凋亡。
AIM: To investigate the effect of hydrochloric ligustrazine injection on expression of induced nitricoxide synthase (iNOS) and apoptosis in rats with spinal cord injury (SCI).
METHODS: The experiment was completed at the Laboratory of Orthopaedics, Union Hospital, Tongji Medical Callege, Huazhong University of Science and Technology from July to November 2005. Totally 72 SD rats were used to establish moderate injured models with modified Allen's method and divided randomly into injured group and ligustrazine group. Rats in the two groups were treated with saline and ligustrazine respectively, and sacrificed at 8-hour, 1-day, 3-day, 1-week, 2-week and 3- week time points after injury. Pathological changes of spinal cord tissue were observed wih haematine-eosin staning, iNOS positive cell was detected with immunohistochemical staining, and apoptosis cells were labeled with in situ end-labeling (ISEL). Histological test, positive cellular rate of iNOS expression and apoptosis index were observed at each time point.
RESULTS: All 72 rats were involved in the result analysis. ① Rats in ligustrazine group could be observed as the followed as compared with those phenomenons in injured group: hematomuellia was decreased obviously, baemorrhage shaped as blot-focus and necrosis were observed in a limited area, neural cellular swelling was mild, edema of tissue was mild and vacuolation was little. ② Positive cells of iNOS expression were observed both in ligustrazine group and injured group. Neurons, neuroglial cells, vascular endothelial cells and ependymal cells were reached at peak within 1 week. There were significant differences of positive cellular rate among 8- hour, l-day, 3-day, l-week, 2-week and 3-week time points (t=2.234-13.412, P 〈 0.05). ③ Apoptosis cells were observed both in ligustrazine group and injured group, and reached at peak within 1 week. There were significant differences of apoptosis index among 8-hour, 1-day, 3-day, 1-week, 2-week and 3-week time points (t=2.417-3.689, P 〈 0,05).
CONCLUSION: Ligustrazine injection can inhibit iNOS expression and apoptosis in rats after spinal cord injury.
