摘要
研究了DBM-偶氮胂(DBM—ASA)与血清白蛋白(BSA)的反应。在室温下,于pH2.9的Britton—Robinson缓冲溶液中及乳化剂OP存在下,DBM—ASA与血清白蛋白复合物最大吸收峰位于530nm处,复合物表观摩尔吸光系数εmax=5.73×10^3L/(mol·cm),BSA在浓度10.0~75.0mg/L范围内呈良好线性关系。加入乳化剂OP可显著提高体系灵敏度与稳定性。加入少量乙醇能提高反应灵敏度。用本法测定了人血清样品中的蛋白质含量,结果满意。
The reaction between DBM-arsenazo and bovine serum albumin was studied. In a Britton-Robinson buffer solution at pH 2.9 and in the presence of emulgent OP, DBM-arsenazo and bovine serum albumin form a compound with a maximum absorption wavelength at 530 nm. The apparent molar absorptivity of the compound is εmax = 5.73 × 10^5 L/ (mol'em). The linear range of bovine serum albumin is 10 ×75.0 mg/L. The addition of emulgent OP can obviousl) enhar.ee the sensitivity and stability of the eolour system. The addition of a small amount of alcohol can increase the sensitivity of the eolour system. The protein in human serum was determined by the proposed method with satisfactory results.
