摘要
本文用同位素参入法研究不同月龄大鼠的肝细胞核体外转录活性。结果观察到老年大鼠的肝细胞核体外转录活性较断乳鼠及青年鼠分别下降了47%及42%,而肝组织中总RNA的含量随增龄变化不十分明显;负责rRNA及tRNA的RNA聚合酶Ⅰ+Ⅲ所致的转录活性随增龄明显下降,而负责n.RNA合成的RNA聚合酶Ⅱ所致的转录活性则改变不太明显;RNA合成后从细胞核转运到核外的变化趋势是1~10月龄时上升,11—28月龄时下降。
By means of incorporation of [~3H] UMP, changes in RNA synthesis of liver nuclei were determinedat different ages of male rats. RNA synthesis in rat liver nuclei was found to decrease about 47%between 1 and 26 months, and 42% between 7 and 26 months respectively, but the content of RNA inrat liver tissue did not change obviously with increasing age. The synthesis of rRNA and tRNA by polymerase Ⅰ and Ⅲ decreased about 59% from 1 to 10months, and 38% from 10 to 26 months in rat liver nuclei, on the other hand, the synthesis of mRNAby polymerase Ⅱ deoreased with increasing age, but witkout signifficant difference statisticaliy. The transport of RNA from nuclei to cytoplasm was determined by using rat liver nuclei. Anincrease was found from 1 to 10 months, and a deorease from 10to 26 months.
