复方茯苓甘草汤对慢性低氧大鼠肺内细胞增殖与凋亡的影响 被引量:5

Effects of Fufang Fulin Gancao Decoction on Pulmonary Cell Proliferation and Apoptosis in Chronic Hypoxic Rats

摘要 目的:研究复方茯苓甘草汤对慢性低氧大鼠肺内p53及PCNA蛋白表达及细胞增殖与凋亡的影响。方法:30只雄性SD大鼠随机分为对照组(A组,各组n=10),低氧组(B组)和低氧+复方茯苓甘草汤治疗组(C组)。采用常压间断低氧8h/d,连续21天的方法复制慢性低氧性肺动脉高压模型。观察复方茯苓甘草汤对慢性低氧大鼠肺血管形态学指标、p53及PCNA基因蛋白表达、细胞的增殖与凋亡的影响。结果:①低氧21天后,B组MT%、MA%、PI及p53基因表达较A组均明显升高(P<0.01),VA%、AI较A组明显降低(P<0.01);②C组MT%、MA%、PI及p53基因表达与B组相比明显降低,差别有显著意义(P<0.01或P<0.05)。VA%、AI与B组相比明显升高,差别有显著意义(P<0.01);C组除VA%、AI及p53基因表达与A组比差别有意义(P>0.05)外,其它指标与A组比较差别均无意义(P<0.01或P<0.05)。结论:复方茯苓甘草汤可抑制肺血管重建,其机理可能与抑制p53及PCNA基因的表达,导致细胞增殖被抑制、细胞凋亡被促进等有关。 Objective :To investigate the effect of Fufang Fulin Gancao Decoction on expression of p53 and PCNA, on cell proliferation and apoptosis in chronic hypoxic rats. Methods :30 healthy Spraque -Dawley male rats were randomly divided into three groups: group A, normal control group ( n = 10); group B, chronic hypoxic group ( n = 10); group C, hypoxia plus Fufang Fulin Gancao Decoction treatment group( n = 10). Then the model of chronic hypoxic pulmonary hypertension were established by exposing rats to normobaric hypoxic conditions for 8 hours one day in 3 weeks continually except rats of group A. At last, effects of Fufang Fulin Gancao Decoction on vascular morphologic indexes of pulmonary, expression of P53 and PCNA, cell proliferation and apoptoais in pulmonary tissue of HPH rats. Results :(1)After 21 days,rats of group B in MT%, MA%, PI, expression of p53 are obviously higher than group A ( P 〈 0.01 ), in VA%, M are distinctly higher than group A ( P 〈 0.01 ) ; (2)Rats of group C in MT%, MA%, PI and expression of p53 are obviously decreased than group B( P 〈0.01 or P 〈0.05), in VA%and AI are increased higher than group B( P 〈0.01) ; Except that VA% ,M and expression of p53 have meaning ( P 〈0.01 or P 〈0. 05) , other indexes have no meaning compared with group A( P 〉0.05). Conclusion : Fufang Fulin Gancao Decoction can refrain persistently from the development of pulmonary vascular remodeling efficiently, its mechanism may partly relate to inhibiting the protein expression of 1363 and PCNA, enhancing cell proliferation and decreasing cell apoptosis.
