流体剪切应力对人骨肉瘤成骨样MG-63细胞Cbfα1基因表达的影响 被引量:2

摘要 目的观察流体剪切应力(FSS)对人骨肉瘤成骨样细胞(MG-63)Cbfα1基因表达的影响。方法对MG-63骨肉瘤细胞进行培养,传代60h后,将细胞置于流室系统中进行应力刺激实验。FSS分为0.5Pa和1.5Pa两个水平,每个水平下作用时间分别为15min、30min和60min。提取细胞总RNA,进行反转录PCR,检测细胞内Cbfα1mRNA的表达,并计算与内参照GAPDHmRNA的比值。结果与对照组相比,FSS作用组的Cbfα1 mRNA的表达在30min和60min均显著增强,且在一定的时间范围内(15—60min),Cbfα1 mRNA的表达水平随着作用时间的延长、应力水平的增加而增强,在30min和60min的变化具有显著性意义(P〈0.01)。结论FSS可以促进成骨细胞内Cbfα1的表达。 Objective To investigate the effects of flow shear stress(FSS) on the gene expression of Cbfα1 in human osteosarcoma cells(MG-63). Method Human osteosarcoma cells (MG-63) were cultured 60 h, then the cells were treated with 0.5 Pa or 1.5 Pa FSS in a flow chamber for 15, 30, 60 min, respectively. Total RNA in cells was isolated. Reverse transcription PCR analysis was made to examine the gene expression of Cbfα1. Result Comparing with control group, Cbfα1 mRNA expression increased remarkably from 30 min to 60 min with the treatment of FSS (P 〈 0.01 ). And there was clear difference between the expression of Cbfα1 mRNA after treatments of 0.5 Pa or 1.5 Pa FSS from 30 min to 60 min. Conclusion FSS can significantly increase gene expression of Cbfα1 in human osteosarcoma cells.
