胶质瘤中E2F1、MDM2的表达及其与肿瘤恶性程度的关系 被引量:4

Relationship between E2F1 and MDM2 expressions and the malignancy in glioma

摘要 目的研究E2F1、MDM2在不同级别恶性人脑胶质瘤中表达的水平及其与肿瘤恶性程度的关系。方法共82例胶质细胞瘤标本(I级胶质瘤20例、Ⅱ级胶质瘤20例、Ⅲ级胶质瘤21 例、Ⅳ级胶质瘤21例)进行E2F1、MDM2免疫组化染色。实验结果用统计软件SPSS11.0进行卡方检验。结果 E2F1在低级别(I级、Ⅱ级)胶质瘤中的阳性表达率较高,在高级别(Ⅲ级、Ⅳ级)胶质瘤中较低,两者相比差别有显著性意义(X2=3.919,P<0.05);而MDM2在低级别胶质瘤中阳性表达率较低, 在高级别胶质瘤中较高,两者相比差别有极显著性意义(X2=12.602,P<0.01)。结论 E2F1和MDM2 在胶质瘤中所起的作用不同,前者在胶质瘤的发生发展中可能起了抑癌基因的作用,而后者在其中扮演了癌基因的角色。 Objective To detect the expressions of E2F1 and MDM2 gene in gliomas of different malignancy and to explore the relationship between the expressions and the malignant grades. Methods Immunohistochemical staining of MDM2 and E2F1 was performed in 82 cases ofglioma (grade Ⅰ 20 cases, grade Ⅱ20 cases, grade Ⅲ 21 cases, grade Ⅳ 21 cases). Chi-square test was applied for statistical analysis. Results E2F1 and MDM2 expressions were different in different grades of gliomas. It was obvious that E2F1 expression was mainly in low-grade glioma (grade Ⅰ 65.0% and grade Ⅱ 40%, X^2=3.919,P〈0.05), while MDM2 expression was mainly in high-grade glioma(grade Ⅲ 57.1% and grade Ⅳ 76.2%, X^2=-1 2.602, P〈0.01). Conclusion It might be considered that E2F1 and MDM2 act differently in glioma. The role of E2F1 expression in glioma is similar to that of a tumor suppressor gene, while MDM2 acts as an oncogene.
