摘要
钝顶螺旋藻 Spirulina platensis 类囊体膜经提取后,用 SDS-PAGE 分离得三条叶绿素蛋白复合物区带,按迁移率顺序,它们分别是 CPI,CPa 和游离色素带 FC。CPI 在红区和兰区的吸收峰分别为678nm 和441nm,CPa 在红区和兰区的吸收峰分别为676nm 和440nm。室温下 CPI 荧光发射峰位于686nm,而 CPa 荧光发射峰位于684nm。CPI 分子量为87 000,CPa分子量为49 500。测定了 CPI 的氨基酸组成。CPI 远紫外圆二色谱测定计算结果,CPI 的二级结构中α-螺旋含量为35.8%,β-折叠为19.9%,无规卷曲为44.2%。
When the thylakoid membrane of blue algae Spirulina platensis was extracted and then analysed by SDS-Polyacrylamide gel electrophoresis,three green zones were resolved.The first was the chlorophyll-protein complexes of PSI (CPI),the another was the chlorophyll-protein complexes of PSⅡ (CPa) and the third one was the free chlorophyll zone.The absorption spectrum of CPI had a red maximum at 678nm and a peak in the blue at 441nm.The abaorp- tion spectrum of CPa had a red maximum at 676nm and a peak in the blue at 440nm.The fluorescence emission of CPI had a peak in the red maximum at 648nm at 25℃.The molecular weight of CPI is 87000 and CPa is 49500.In the secondary structure of CPI,there were 35,8%a-helix,19.9% B-pleated sheet and 44.2% random coil.
