摘要
在38~40℃,CO_2与空气比例为40:60和60:40的条件下,利用50%鸡胚提取液,50%鸭胚提取液,M_(100)、F_(12)、羊血清、牛血清、酵母提取物,混合维生素和多种有机和无机物分别组成8组不同成分与浓度的培养液,对达呼尔奥斯特线虫进行了寄生阶段的体外培养试验。在以鸡胚或鸭胚提取物为主,分别加入 M_(100)、牛或羊血清、酵母提取物等成分的培养液中,幼虫经14~17d 培养后,有大约14%~20%通过第三次蜕化达到了第四期。实验结果表明,(1)商品培养基 F_(12),M_(100)对幼虫发育无明显作用;(2)鸡胚或鸭胚提取物对幼虫一定阶段的发育有较好的效果;(3)不同寄生阶段的幼虫要求不同的营养成份和环境条件。
The media used for in vitro cultivation of O.dahurica were com posed of various kinds and different concentrations of serum,embryo extracts and vitamins,and were also used as essential ingredients.Eight roller culture systems were designed for cultivation.Exsheathed L_3 of O.dahurica was cultured in different media with a gas phase of 40%CO_2+air or 60%CO_2+airat 38~40℃. L_3 did not show obvious growing charges in the following media;(1) sheep serum+M_(190),and (2) sheep serum+F_(12).Medium containing chicken embryo extract (or duck embryo extract),calf serum,M_(199),yeast extract,and different kinds of vitamins with or without mixed organic,inorganic compounds supported good growth,14%~20% of L_2 metamorphosed into L_4.
