摘要
目的利用正交实验研究紫菀中紫菀酮的最佳提取条件。方法利用微波辅助加压溶剂提取法提取紫菀中紫菀酮,通过正交实验,考察了微波提取条件(包括溶剂、提取时间、提取次数和料液比)对紫菀中紫菀酮的影响。采用HPLC检测紫菀酮的含量,其中色谱柱:Spherisorb C18(4.6 mm×250 mm,5μm);流动相:乙腈-水(98∶2);流速:1.0 mL.min-1;柱温40℃;检测波长为200 nm。结果溶剂使用体积分数为80%乙醇,提取时间为30 min,提取次数为3次,料液比为1∶20时,提取率最佳。结论采用本实验方法提取紫菀中紫菀酮具有效率高,时间短,所需温度低以及绿色环保等优点。
OBJECTIVE The optimum extraction conditions of shionone from herbs Tatarian aster root were studied by means of oahogonal test and microwave-assisted and pressurized solvent extraction technique. METHODS The influence of some operation conditions, such as ethanol content, microwave irradiation time, extraction times and the ratio of sample weight to solvent volume, on the extraction yield of shionone have been studied by using orthogonal test and multivariate statistics. The contents of shionone were determined by HPLC. The separation was performed on Spherisorb C18 column (4.6 mm× 250 mm,5 μm) with acetonitrile-water (98:2) as mobile phase. The flow rate was 1.0 mL·min^-1, the column temperature was 40 ℃ and the wavelength of UV detector was 200 nm. RESULTS The highest extraction yield was obtained when extraction solvent was 80% ethanol, microwave inadiation times is 30 minutes, the samples were extracted 3 times and the ratio of sample weight to solvent volume was 1 : 20. CONCLUSION The method had higher extraction efficiency as well as shorter time and lower temperature, was green and no pollution.
