冰冻红细胞洗涤方法的比较研究 被引量:8

摘要 目的比较传统的大型离心机方法与新型透析式洗涤机方法洗涤冰冻犬红细胞的输注效果,评价新型透析式洗涤机的应用价值。方法采用动物实验的方法,观察犬红细胞冰冻保存洗涤后回输体内24h回收率及安全性。取犬全血200ml,分离红细胞进行甘油化保存,1周后解冻犬红细胞,分别经两种方法洗涤后自体回输,取脏器进行组织切片和HE染色。结果两种方法洗涤效果的差异无显著性,两种洗涤方法处理的冰冻红细胞回输犬体内后24h回收率均>90%;回输72h后解剖脏器观察,两组动物脏器均未见异常。结论洗涤的冰冻犬红细胞回输实验动物体内后代谢稳定,对犬脏器无副作用,同时新型透析式洗涤机具有缩短洗涤时间、自动化全封闭式的特点,适宜于临床应用。 Objective To compare two methods,the large centrifuge and the new type of washing machine,and to evaluate the application of new type of washing machine based on dialysis. Methods Researches have been done on canine model. 200 ml blood were taken from every canine, and red blood cells were separated and preserved with glycerol. The glycerolized canine red blood cells were thawed and washed in two different ways and auto-transfused to the canine. The remainder of the washed canine red blood cells after 24 h of metabolism was detected to evaluate the life time limit of the washed canine red blood cells. The animal was killed and autopsied after 72 h of auto-transfusion. Results There was no significant difference between the two washing methods. The 24 h recovery rate of the red blood cells washed with two methods were both above 90%. The histology of the organs were normal without any pathological changes. Conclusion The washed frozen red blood cells transfused to the animals were metabolized steadily. No side effect was found in the organs of the canine recipient. At the same time, it took shorter time to complete washing using the new type of washing machine. The characteristics of automatization and close also make the machine feasible to apply in the clinic.
