摘要
目的:测定人血浆中阿昔洛韦的浓度。方法:采用高效液相色谱荧光法检测,色谱柱为ODS-3(DIKMA150mm×4.6mm,5μm),流动相为0.02mol·L-1高氯酸-甲醇(450∶8),流速为1.0mL·min-1,检测波长为λex=260nm,λem=375nm。结果:阿昔洛韦的提取回收率大于96%,方法回收率为(98.4±3.7)%,日内RSD≤5.7%,日间RSD≤6.6%。结论:本方法准确可靠,可用于阿昔洛韦体内血药浓度测定。
OBJECTIVE To determine the content of acyclovir in human plasma. METHODS The column was ODS-3(DIKMA 150 mm×4. 6 mm,5μm). The mobile phase consisted of 0.02 mL·min^-1 perchlorie acid: methanol(450:8). The flow rate was 1.0 mL·min^-1. The detection wavelengths were λex = 200 nm and λem = 375 nm, respectively. RESULTS The extraction recovery was more than 96% and the recovery of the essay was ( 98. 4 ± 3.7)%. The relative standard deviation(RSD) of intra-day and inter-days were less than 5.7% and 6. 6%, respectively. CONCLUSION The method is accurate and reliable. It can be applied for the determination of acyclovir in human plasma.
