摘要
吗氯贝胺系新型可逆性。选择性单胺氧化酶Ａ型抑制剂。本文首次在国内建立测定人血清中吗氯贝胺的ＨＰＬＣ法，并报告部分病例血清标本的测定结果。以甲氧氯普胺作内标，在碱性条件下用乙醚提取。流动相为乙腈：０．０２３ｍｏ１／Ｌ四甲基乙二胺溶液（２０：８０，Ｖ／Ｖ，ｐＨ６．０）。用紫外２４０ｎｍ测定。吗氯贝胺线性范围为０－４０００μｇ／Ｌ，最低检出限４μｇ／Ｌ。吗氯贝胺的绝对回收率大于９０％，日内变异为１．６－３．２％，日间变异为２．４－５．３％。应用本法对服用吗氯贝胺治疗的１８例抑郁症病人的５４份血清标本进行测定，结果表明本方法与国外同类方法具有可比性。
Moclobemide is a new type of reversible and selective inhibitor of the enzyme monoamine oxidase subtype A(RIMAs.).A specific and sensitive HPLC method for the determination of Moclobemide in human serum,using metoclopramide as the internal standard,is first presented.The mobile phase is consisted of acetonittile and 0.023 mol/L tetrarnethy ethylemediamine solution(20:80,v/v,pH 6.0).Basified samples are extracted with ether and are measured with UV detector at 240 nm.The relative recovery of moclobemide is more than 90%.Excellent linearity is obtained over the range of 0 to 4000μg/L,and the limit of detection is 4μg/L.Within day and day-to-day coefficients of variation are 1.6-3.2% and 2.4-5.3% respectively.54 serum samples,offered by 18 depression patients who were treated with moclobemide,were determined by the procedure.The result indicates that our method has the similar effect compared with such method used abroad.
