摘要
目的观察切除大鼠甲状旁腺后,牙本质形成的改变,体内研究甲状旁腺激素(parathyroid hormone,PTH)对成牙本质细胞分化和矿化的影响。方法采用动物实验和组织学切片的方法,观察甲状旁腺自体种植组和甲状旁腺摘除组与对照组相比的牙本质形成变化情况。结果对照组和甲状旁腺自体种植组成牙本质细胞和前期牙本质无明显改变;甲状旁腺摘除组前期牙本质小管增宽,前期牙本质和牙本质层出现矿化缺乏裂隙,成牙本质细胞沿前期牙本质内侧排列,形态无明显变化。甲状旁腺摘除组中,血钙含量高的大鼠前期牙本质和牙本质层的形态反而接近于对照组。结论PTH缺乏导致的血钙降低可影响牙本质形成,结合体外实验结果,提示PTH通过激素本身及调节血液钙含量影响成牙本质细胞分化和牙本质形成。
Objective:To determine the effects of parathyroid hormone( PTH ) upon dentinogenesis in rat tooth. Methods : 15 SD rats aged 42 d were evenly divided into 3 groups. In the groups of control, PIT and PTX, shamoperatoin, parathyroid autotransplantation and parathyroidectomy were conducted respectively. After operation serum Ca^2+ was weekly measured with methyhhymol method. 30 d after operation dentinogenesis was observed with HE staining and light microscopy. Results:30 d after operation,serum Ca^2+ concentration in PTX group was lower than that in control and PIT groups(P 〈0.05). In control and PTT groups the morphology of dentin was similar. In PTX group, enlargement of dentin tuble in the predentin, alterations in the predentin appearance and the presence of mineralization defects were observed. Conclusion..PTH deficiency may result in dentinogenesis defect.
