气相色谱法检测健康人血清游离脂肪酸 被引量:6

摘要 目的测定健康人血清中7种游离脂肪酸(FFA)含量。方法采用气相色谱法(GC)测定血清FFA。血清样品前处理:以正十七酸为内标,用硫酸/甲醇溶液对血清中FFA进行甲酯化处理。结果α亚麻酸、花生四烯酸、二十碳五烯酸和二十二碳六烯酸的线性范围在2~300μg/mL,硬脂酸、油酸和亚油酸的在50~1000μg/mL(r>0.999)。方法回收率良好(95.65%~104.88%)。各种脂肪酸的日内变异系数为0.36%~2.13%(n=5),日间变异系数为0.82%~3.36%(n=5)。结论气相色谱法测定血清FFA方法灵敏、准确,可用于临床。 Objective To establish method for the determination of 7 free fatty acids in human serum. Methods Free fatty acids were detected by gas chromatography(GC). The extracted lipids were methylesterified by H2SO4/methanol. Heptadecanoic acid(C17 : 0) was used as internal standard. Results The linearity of C18 : 3, C20 : 4, C20 : 5 and C22 : 6 ranged from 2-300 μg/mL, and linearity of C18 = 0, C18 = 1 and C18 : 2 ranged from 50-1 000 μg/mL(r〉0. 999). The recovery rate was 97. 65% - 104.88 %. The intra-day variation coefficient was less than 2. 13 % and 3. 36 %, respectively. Conclusion This method proved to be sensitive, reproducible, and applicable for the determination of free fatty acid in serum.
