摘要
目的探讨B7H1在混合皮肤移植中自体角朊细胞诱导的局部免疫耐受中的作用。方法在加入与不加入自体角朊细胞的实验系统中,利用混合表皮淋巴细胞培养(MELC)体外模拟系统,检测角朊细胞表达B7H1的情况,并观察抗B7H1和PD1单抗对自体角朊细胞诱导的免疫抑制的影响情况。结果在引入自体角朊细胞的MELC体系中,刚分离的角朊细胞并不表达B7H1,但在4h开始有微弱的表达,而>8h就有较强的表达;而未引入自体角阮细胞的MELC体系中,则始终未检测到B7H1的表达。而且自体角朊细胞对自体淋巴细胞对异体表皮细胞的同种异体增殖反应的抑制作用能被抗B7H1和PD1的单抗阻断。结论混合皮肤移植中,自体角朊细胞可能是通过B7H1提供共刺激信号,激活Th2亚群,遏止Th1亚群,从而抑制后者介导的细胞免疫。
Objective To explore the effects of B7-H1 molecule in local immuno-anergy induced by atuo-keratinocytes(Auto-Kc) in the intermingled skin grafting. Methods B7-H1 was detected to be expressed on keratinocytes by PT-PCR. The change of down regulatory ability of Atuo-Keratinocytes was observed when added with mAb against PD-1 and B7-H1 molecule in mixed lymphocyte culture (MELC) system. Results B7-H1 cDNA began to be amplified specifically on 4 h after initiating of the MELC syetem with auto-Kc(La+Eb+Kca). On 8-16 h,the expression was increased and reached its maximum. Anti-B7-H1 and PD-1 mAb almost able to block all of auto-Kc's ability of inducing suppression. Conclusion Auto-Kc activated the Th2 cells by indirect recognition pathway, in which B7-H1 molecule provided costimulaory signal, and then inhibited the proliferation of the Thl cells, leading to a local immuno-anergy for allo-rejection.
