摘要
应用人肝癌SMMC-7721细胞系和大鼠肝原代培养细胞,利用[3H]TdR掺入的方法观察从人胎肝中提取的肝细胞生长刺激物质(HSS)对肝源性细胞NDA合成的影响。证明HSS对此两种细胞的DNA合成均有显著的促进作用。应用HSS治疗D-氨基半乳糖引起的大鼠肝坏死模型,结果使大鼠的血清谷丙转氨酶有非常显著的下降。本研究进一步证明肝细胞生长刺激因子在人胎肝中存在,并且对肝源性细胞有特异的促生长作用。
Hepatic stimulator substance(HSS) was extracted from human fetal liver.It was tested for growth stimulatory activity in two kinds of hepatoma cells.The result showed that HSS had a remarkable effect in [3H]TdR incorporation into SMMC-7721 cell DNA and rat liver primary cultivate cell DNA.Sprague-Dawley rats were injected with D-galactosamine to induce acute liver failure.HSS suppressed the elevation of serum ALT induced by D-galactosamine in a dose-dependent manner.These experiments prove that HSS exists in human fetal liver and promotes proliferation of liver cells
