狗脑抗吗啡活性肽“74a”的分离纯化方法及其鉴定

摘要 本实验报道从正常狗脑中获得的第三个具有明显拮抗吗啡镇痛效应的肽,暂定为“74a”肽。所得纯品经等电聚焦电泳和SDS电泳鉴定均为一条带,分子量约10,000道尔顿,等电点pH7.5左右,N-末端35个氨基酸残基序列已经测定,经检索未见文献报导,是一个新肽。 A pare peptide from dog brain is obtained. The whole dog brain was homogenized and extracted with 0.1M acetic acid. The extract was centrif aged at 15000g for 30 min. The supernatant was lyophilized and the residue was dissolyed in 0.1M acetic acid and was allowed to go through Sephadex G-50 column (twice), CM22 cellulose and Sp-Sephadex C-25 ion exchange column. The sample obtained was applied to a HPLC (0.5×30cm) with cross linking (3-10μ). The sample was called 74a peptide, and shows only one band on SDS-polyacrymid gel electrophoresis or isoelectric focusing. Its isoelectric point was around pH 7.5, and M. W. was about 10000 dalton. The sequince of the N-terminal 35 amino acid residue has determined. The 74a peptide show of a sinificant anti-morphine activity,This is a new peptide.
