摘要
研究了海藻酸钙包埋法固定化技术在甾体化合物的生物转化中的应用.利用固定化Penicillium raistrickii细胞实现对左旋乙基甾烯双酮(13-ethyl-estr-4-ene-3,17-dione)的15α位羟基化作用,合成了重要的药物中间体15α-羟基左旋乙基甾烯双酮.优化了固定化孢子浓度,固定化孢子接种量,投料浓度,投料时间,转化时间等转化条件.结果表明:在摇瓶发酵生物转化中,固定化Penicillium raistrickii细胞可重复利用5次而转化能力没有明显下降.
15α-hydroxylation of 13 ethyl-estr-4-ene-3, 17-dione using immobilized Penicillium raistrickii entrapped in calcium alginate gel was studied. The conversion conditions, which affected hydroxylation such as substrate concentration, the time to add substrate, conversion duration, were investigated. The results show that, the immobilized Penicilllum raistrickii can be utilized repeatedly five times with no obvious decline of bydroxylation ability.
