摘要
目的:观察大枣多糖对血虚大鼠全血细胞及红细胞膜ATP酶活力的影响,分析大枣的补气血作用的可能途径。方法:实验于2002-02/06在河南中医学院动物实验中心完成。①选用Wistar大鼠60只,体质量170~190g,雄性。随机选用50只采用以下方法造成气血双虚模型:分别于实验第1,3,5,7,9天,尾部放血1mL/180g,于第2天腹腔注射环磷酰胺生理盐水溶液40mg/kg,于第4,6,8,10,12天腹腔注射环磷酰胺20mg/kg,造气血双虚模型;其余10只大鼠为空白对照组(仅注射同体积生理盐水)。②将50只气血双虚模型大鼠随机分为5组:大、中、小剂量大枣多糖组[灌胃大枣多糖溶液,由河南中医学院药物化学学科提供。本品为鼠李科植物枣干燥成熟果实的提取精制物(经乙醇回流脱脂、水提取、浓缩、醇沉、除蛋白、透析、醇沉、真空干燥等程序,得大枣多糖)溶液剂量为200,100,50mg/kg,灌胃体积为10mL/kg],当归补血口服液组[灌胃当归补血口服液(郑州市协和制药厂生产,剂量为2次/d,10mL/次,批号010301)3.3mL/kg,原药液稀释1.5倍]及模型组(灌胃等体积生理盐水)。空白对照组仅灌同体积生理盐水。每天给药1次,连续给药14d。③于第14天给药后2h,用库尔特细胞全自动分析仪测定血中红细胞计数、白细胞计数、血红蛋白和血小板计数。ATP酶测定用定磷法,按南京建成生物工程研究所提供的ATP酶试剂盒说明书测定红细胞Na+-K+-ATP酶和Ca2-Mg2-ATP酶活性。④计量资料组间比较采用t检验,方差不齐者采用t’检验。结果:大鼠60只均进入结果分析。①空白对照组、各剂量大枣多糖组、归补血口服液组大鼠血红细胞计数、白细胞计数、血小板计数及血红蛋白含量明显高于模型组(P<0.05~0.01)。②空白对照组、各剂量大枣多糖组、归补血口服液组大鼠红细胞Na+-K+-ATP酶、Mg2+-ATP酶、Ca2+-Mg2+-ATP酶活力明显高于模型组(P<0.05~0.01)。空白对照组,大、中剂量大枣多糖组、归补血口服液组大鼠红细胞Ca2+-ATP酶活力明显高于模型组(P<0.05),小剂量大枣多糖组大鼠Ca2+-ATP酶活力与模型组相近。结论:大枣多糖可以改善气血双虚大鼠的造血功能和红细胞能量代谢,从而起到补血作用。
AIM: To observe the effect of fructus jujubae polysaccharide (FJP) on complete blood cells and activity of ATPase in rats with blood deficiency, so as to explore the probable methods of fructus jujubae on nourishing qi and blood.
METHODS: The experiment was accomplished at Animal Experiment Center of Henan College of Traditional Chinese Medicine between February and June 2002. ①Fifty of Sixty male Wistar rats with 170-190 g body mass were randomly selected to establish the both qi and blood deficiency model by followed method: The model was established through tail bloodletting 1 mL/180 g on the 1^st, 3^nd, 5^th, 7^th and 9^th days, and intraperitoneal injection of cyclophosphamide normal saline 40 mg/kg on the 2^nd day and cyclophosphamide 20 mg/kg on the 4^th, 6^th, 8^th, 10^th and 12^th days; the left 10 rats were served as the blank control group, which were only injected with the same volume of normal saline. ②The 50 rats in the model group were divided into 5 groups: Large, middle and small-dose FJP groups [The FJP was provided by the medicinal chemistry subject of Henan College of Traditional Chinese Medicine. It was the inclusion from ripe and dry fruitage of Ziziphus jujuba Mill (circumfluence degrease by ethanol, distill by using water, inspissation, sedimentation by ethanol, getting rid of protein, dialysis, sedimentation by ethanol, torrefaction in vacuum and So on to get the FJP) with a dose of 200, 100, 50 mg/kg, and the volume of intragastric infusion was 10 mL/kg], Danggui Buxue Koufuye group [Danggui Buxue Konfuye was given intragastric infusion (produced by the Xiehe Pharmacy Factory in Zhengzhou, once 10 mL, twice every day, Lot No. 010301) 3.3 mL/kg with dilution of original liquor by 1.5 times], and model group (intragastric infusion with the same volume of saline). The rats in the blank control group were only injected with the same volume of normal saline. After establishment of models, the rats were administrated once daily for 14 days.③Mter 2 hours of administration on the 14^th day, the count of red blood cells (RBC), white blood cells (WBC), hemoglobin (Hb) and platelet (PLT) in the blood were measured with the Coulter Automatic Cell Analyzer. ATPase was detected with quantitation of phosphorus method: the activity of Na^+-K^+-ATPase, Mg^2+ -ATPase, Ca^2+ - ATPase and Ca^2+ -Mg^2+ -ATPase in RBC were measured according to the introduction of ATPase kit provided by Nanjing Jiancheng Bioengineering Institute. ④The t-test was used in the comparison of measurement data among groups, and the t'-test was used in analysis of heterogeneity variance.
RESULTS: All the 60 animals were involved into the result analysis. ① Compared with the model group, the level of RBC, WBC, PLT and Hb in the blank control group, large, middle, small-dose FJP groups and the Danggui Buxue Koufuye group were remarkably higher (P 〈 0.05-0.01). ②The activity of Na^+-K^+-ATPase, Mg^2+-ATPase and Ca^2+ -Mg^2+ -ATPase of RBC in the blank control group, large, middle, small-dose FJP groups and the Danggui Buxue Koufuye group were obviously higher than that in the model group (P 〈 0.05-0.01). The activity of Ca^2+ -ATPase of RBC in the blank control group, large, middle-dose FJP groups and the Danggui Buxue Koufuye group was obviously higher than that in the model group (P 〈 0.05), while the small-dose FJP group was close to the model group,
CONCLUSION: FJP can enhance the hematopoietic function and energy metabolism of RBC, which plays a role in nourishing blood,
