摘要
巨桉是造纸和人造板的良好材料,其组织培养微繁殖体系已经研究成功。取优树主干基部萌芽或扦插苗1~1.5cm长的茎段做为外植体,外植体和增殖培养在附加有BAP和NAA的MW培养基上进行。培养条件为26±2℃。日光灯光照10h·d^-1.增殖培养采用BAP0.1—0.5mg·L^-1和NAA0.01~0.1mg·L^-1.离体的茎芽适合在B2附加0.5mg·L^-1IBA生根。小苗移栽红心土上,在简易温室成活率高达93%。
A mieropropagation system was developed for Eucalyptus grandis Hill ex Maiden, which is widely used for pulp and artificial boards. Shoots of 1 - 1.5 cm long from the base of plus tree trunks or cutting seedlings served as explants. The explants and shoot multiplication cultures were cultured on MW medium supplemented with combinations of BAP and NAA. Cultures were maintained at 26 + 212 and exposed to 10-h photoperiods of cool-white fluorescent light. Shoot multiplication cultures were maintained by BAP at 0.1- 0.5 mg· L^-1 and combined with 0.01-0.1 mg· L^-1 NAA. In-vitro-produced shoots were suitable to root on 0.5 mg· L^-1 IBA supplemented to medium 132. Plantlets survived in red soil and exhibited high survivability (more than 93 % ) under simple greenhouse conditions.
