摘要
目的观察粒细胞集落刺激因子(G-CSF)及胸腺肽α1刺激后,人单核细胞株(THP-1)对内毒素刺激的反应及对Toll样受体4mRNA表达的影响。方法实验采用人THP-1细胞,以10、100及1 000ng/mL的G-CSF及胸腺肽α1分别刺激4h或24h。继而以10ng/mL内毒素刺激45min。以RT-PCR法扩增Toll样受体4(TLR4)mRNA。ELISA法测定肿瘤坏死因子-α(TNF-α)以表示细胞对内毒素的反应。结果细胞经G-CSF及胸腺肽α1刺激后TLR4表达增加,细胞分泌TNF-α上升。随着G-CSF及胸腺肽α1刺激浓度的增加,TNF-α表达随之增加;刺激24h与4h比较,TNF-α表达也有增加。结论G-CSF及胸腺肽α1可刺激THP-1细胞TLR4mRNA表达,增加细胞对内毒素的反应,增加程度与G-CSF及胸腺肽α1刺激浓度及刺激时间呈正性相关。
Purpose To study THP-1 cell line reactivity to endotoxin stimulation and the expression of Toll-like receptor 4 (TLR4) after thymosin α1 and granulocyte colony-stimulating factor(G-CSF) stimulation. Methods Human THP-1 cell line were stimulated by thymosin α1 and G-CSF with 10, 100 and 1 000 ng/mL for 4 h or 24 h. The cells were then stimulated with endotoxin for 45 min. TLR4 genes were amplified using RT-PCR and TNF-α was detected with ELISA. Results TLR4 mRNA and TNF-α were up-regulated in THP-1 cells after stimulated with thymosin α1 and G-CSF. With the increasing of the concentration of thymosin α1 and G-CSF, the expression of TNF-α also increased; the level of TNF-α was higher when stimulated for 24 h than that of 4 h. Conclusions The expression of TLR4 mRNA and TNF-α was up-regulated in THP-1 cell line by the stimulation of thymosin α1 and G-CSF and had a positive relation to the drug concentrations and stimulating time.
