摘要 目的探讨表皮生长因子(EGF)促分裂原活化蛋白激酶(MAPK)信号通路对雄激素非依赖性人前列腺癌细胞系PC-3细胞黏附、迁移、侵袭作用中的影响。方法应用细胞黏附、迁移、侵袭能力测定EGF和MAPK信号通路抑制剂对PC-3转移表型的作用。结果EGF可促进PC-3对纤连蛋白Fn的黏附、迁移和侵袭作用,而MAPK信号通路抑制剂PD98059则具有抑制的作用。结论EGF可能主要通过MAPK信号通路,从而促进PC-3细胞的的黏附、迁移和侵袭能力。 Purpose To investigate the effect of MAPK signal pathway in the EGF's regulation on meastatic phenotypes of prostate cancer cell line PC-3. Methods By using cell adhesion, migration and invasion ability assay, the effects of EGF and PD98058 (an inhibitor of MAPK kinase on metastatic phenotypes in PC-3) were investigated. Results EGF significantly promoted PC-3 cell the adhesion, migration and invasion ability to fibronectin, and PD98058 had the contrary function. Conclusions EGF might promote the metastatic ability mainly by activating MAPK signal pathway in PC-3 cells.
