肝细胞癌组织中PTEN蛋白表达的意义

摘要 目的:探讨肝癌组织中抑癌基因PTEN蛋白表达的意义。方法:应用免疫组织化学S-P法检测37例肝细胞癌及相应的癌旁组织与6例正常肝组织中PTEN蛋白的表达情况,分析HCC中PTEN蛋白表达与各临床病理特征关系。结果:PTEN蛋白在正常肝组织及HCC癌旁组织中表达高于癌组织,差异有显著性(P<0.05),但正常肝组织与癌旁组织表达无显著性差异(P>0.05)。分化程度高的HCC中PTEN表达高于低分化者,ChildA级高于B和C级,差异有显著性(P<0.05);无静脉浸润者高于有静脉浸润者,差异有显著性(P<0.05);PTEN蛋白表达在性别、年龄、有无包膜、大体类型及肿瘤大小间无明显差异(P>0.05)。结论:肝癌组织中PTEN蛋白阴性表达率较高,PTEN可作为检测肝癌生物学特征的一种分子标记。 Objective:To investigate the expression of tumor suppressor gene PTEN protein in HCC tissues and its clinicopathological significance. Methods:The expression of PTEN protein was detected by immunohistochemistry(S-P Method ) in 37 cases with HCC and their corresponding adjacent tissues,6 cases with normal liver tissues. The relationship between the expression of PTEN protein in HCC tissues and all types of clinicopathological characteristics was analysed. Results:The expression of PTEN protein was significantly stronger in the pericarcinomatous liver tissues and the normal liver tissues than in HCC tissues (P〈0.05),but no significant difference was found between normal liver tissues and the pericarcinomatous liver tissues. HCC with higher differentiation, no vain invasion and Child A showed remarkably stronger expression (P〈0.05);The expression of PTEN protein in HCC was not related toy patient's age, sex, tumor size, integral capsule and tumor modality(P〉0.05). Conclusions:There was a higher incidence of negative expression of PTEN protein in HCC, which indicates that PTEN gene inactivation may play an important role in the genesis and development of HCC. PTEN Protein might serve as a molecular marker to detect biological characteristics of HCC.
